番茄红素抗活性氧作用机制
he Role of Lycopene against ROS
番茄红素是一种脂溶性类胡萝卜素,具有很强的抗氧化活性而呈现出有益健康的效应。活性氧(ROS)是细胞内信号调控的第二信使,通过改变细胞的氧化还原状态参与多种反应,在细胞增殖、分化、凋亡等调控中起着重要的作用。在正常情况下,生物体内的抗氧化体系可以维持ROS的代谢平衡,保证细胞正常的生命活动;但外界环境刺激下(如 UV,热,药物等),快速增加的ROS会引起机体氧化应激和过氧化损伤,引发多种器官功能异常或组织病变。研究发现番茄红素不仅能直接清除ROS,而且还能调控ROS的产生,介导ROS作用的信号传导途径,抑制或逆转由ROS引起的损伤。为更好地了解和认识番茄红素抗氧化作用及其调控氧化还原信号途径的机制,本文就番茄红素调控的氧化还原信号途径的分子靶点、酶的活性和表达的影响以及调控蛋白的活性和转录因子等方面的研究进展进行了简要的综述。
Lycopene, a water-insoluble carotenoid pigment, has been associated with the health benefits by virtue of its strong antioxidant activity. Reactive oxygen species (ROS) as second messengers have important roles in in cell proliferation, differentiation and apoptosis as regulatory mediators in signaling processes. Normally, ROS levels are controlled by an antioxidant system. However during times of environment stress (e.g., UV, heat exposure, drug), ROS levels increasing dramatically can result in oxidative damage and oxidative stress, and often induce irreversible functional alterations or even destruction. Evidence is accumulating to suggest that lycopene can not only scavenge ROS directly, but also may act as a modulator of redox signaling and DNA repair. In this review, we report the available evidence on a role of lycopene as a redox agent against ROS, especially on lycopene modulation of redox-sensitive targets, enzyme, transcription factors.
