以小麦-叶锈菌为研究体系,探讨细胞自噬相关基因TaATG8在由真菌诱导寄主细胞发生过敏性坏死过程中的作用。运用实时定量PCR、免疫印迹技术验证并呈现自噬参与过敏性坏死的模式。结果显示,接种后36h,该基因在不亲和组合中RNA水平和蛋白水平的表达明显高于亲和组合,初步表明该基因参与了小麦抵抗叶锈菌侵染的防卫反应表达过程。利用病毒诱导的基因沉默技术(VIGS)沉默TaATG8后,叶锈菌侵染诱发的过敏性坏死出现了扩展现象,表明自噬在叶锈菌侵染小麦诱发的过敏性反应中可能起到负调控作用。
Based on the wheat-leaf rust fungus interaction system, the roles of autophagy related gene TaATG8 during the hypersensitive reaction induced by fungal infection was discussed. RT-qPCR and western-blotting were applied to verify and present the mode of autophagy in this process. The results showed that the expression of gene in the RNA and protein level were significantly higher in the incompatible combination than those in the compatible combination at 36h after inoculation. This preliminarily indicated TaATG8 had participated the expression of defense response in the process of wheat resisting the fungus infection . After virus-induced gene silencing(VIGS) was used to silence TaATG8, hypersensitive reaction which induced by leaf rust infection began to expand, which indicated that autophagy may play a negative role in the hypersensitive reaction .
