复方阿胶浆对移植性lewis肺癌组织凋亡蛋白表达影响
Effects of Compound E-Jiao Slurry (CEJS) on apoptotic proteins in Mice with Lewis Lung Carcinoma
目的:观察复方阿胶浆联合化疗对移植性lewis肺癌组织凋亡蛋白表达的影响。方法:构建lewis肺癌移植瘤模型,随机分为模型对照、环磷酰胺、复方阿胶浆、复方阿胶浆(大、中、小剂量)联合环磷酰胺共6组。给药结束后,取瘤组织石蜡包埋、切片,免疫组化法检测Bcl-2/Bax蛋白表达,图像分析软件测量平均光密度。结果:实验各组均能下调Bcl-2蛋白表达水平,上调Bax蛋白表达,降低Bcl-2/Bax比值,与模型对照组比较,有统计学意义(p<0.01~0.05);与环磷酰胺组比,复方阿胶浆大剂量联合环磷酰胺可下调Bcl-2表达,复方阿胶浆中剂量联合环磷酰胺能够上调Bax表达,复方阿胶浆大、中剂量联合环磷酰胺可下调Bcl-2/Bax比值,均有统计学意义(p<0.01~0.05)。结论:复方阿胶浆可能通过影响Bcl-2/Bax蛋白表达,从而诱导肺癌细胞凋亡,伍用化疗可能增强化疗药物的抗凋亡效应。
Objective: To observe the effects of CEJS combined with chemotherapy drugs on apoptotic proteins in lewis lung carcinoma xenografts. Methods: Tumor xenografts models were prepared and randomly divided into six groups including the control group, CTX, CEJS, high-dose CEJS joint CTX, middle-dose CEJS joint CTX and low-dose CEJS joint CTX group. Then we performed paraffin imbedding and section. The expression of Bcl-2/Bax were determined by immunohistochemistry. The average optical density were evaluated by image analysis software. Results: The expression level of Bcl-2 could be significantly down-regulated and the level of Bax could be significantly up-regulated with Bcl-2/Bax lower in every experimental group, compared with the control group(p<0.01~0.05). Compared with the CTX group, the average optical density of Bcl-2 were obviously lower(p<0.05)in high-dose CEJS joint CTX group. The average optical density of Bax were significantly higher(p<0.05)in middle-dose CEJS joint CTX group and Bcl-2/Bax were lower(p<0.01)in high or middle-dose CEJS joint CTX group, compared with the CTX group Conclusion: CEJS could induce the cell apoptosis of lung cancer by influencing the expression of Bcl-2/Bax proteins and enhance the anti-apoptotic effect caused by chemotherapy drugs.
