蚕素Bombyxin A-4在家蚕杆状病毒表达系统中的表达与功能研究
Expression of BombyxinA-4 in Bombyx mori Baculovirus Expression System and Study of its Function
蚕素是家蚕体内一种5 kD大小的小肽,同人的胰岛素具有40%左右的序列同源性,被确定为胰岛素家族的一种。本研究首先根据蚕素原BombyxinA-4的ORF序列,以重叠延伸PCR(SOE-PCR)技术体外延伸获得B链和A链重组的目的基因BA,构建原核表达质粒pGEX4T-1-BA,转化大肠杆菌E.coli Rosetta-gami(DE3)pLysS表达融合蛋白,纯化后免疫新西兰雄兔获得检测抗体。再从家蚕组织cDNA文库中扩增获得蚕素BombyxinA-4的ORF序列,利用 Bac-to-Bac系统,将目的基因重组至Bacmid上,转染家蚕BmN细胞,获得重组病毒vBmBombyxinA-4,再将该重组病毒接种于家蚕蛹,待大部分家蚕蛹出现明显病毒感染症状后,收集并进行Western blotting检测。结果表明蚕素BombyxinA-4在家蚕蛹中得到表达,该表达产物能够促进家蚕卵巢细胞BmN的生长,同时将正常的家蚕蛹和感染重组病毒的家蚕蛹分别治成粉剂,在高血糖小鼠模型上进行了降血糖作用的研究,结果发现感染重组病毒的家蚕蛹比正常家蚕蛹的降血糖作用明显,为开发新的降血糖类的药品和保健品奠定了基础。
Bombyxin is a 5 kDa brain peptide of the silkmoth Bonbyx nori, whose amino acid sequences have 40% identity with human insulin , belonging to the insulin peptide family. In this study, the target BA gene recombianted with B Chain and A Chain was obtained by SOE-PCR according to BombyxinA-4 ORF. The target BA gene was cloned into pGEX4T-1 to constuct expression vector pGEX4T-1-BA, then recombiant protein was expressed in E.coli Rosetta-gami(DE3)pLysS and purified, the purified protein immunize male New Zealand rabbit and the obtained antibody was used for detection. Then a Bombyxin protein gene (BombyxinA-4) sequence was obtained from silkworm cDNA library constructed by our laboratory. The target gene was recombined to Bacmid by Bac-to-Bac baculovirus expression system, transfected into BmN cells to generate recombinant baculovirus vBmBombyxinA-4. Then the silkworm pupae was infected with vBmBombyxinA-4. Results of Western blotting showed that the BombyxinA-4 could be expressed in silkworm pupa.The expression product can promote the growth of BmN cells. The normal pupa and the pupa infected by recombinant virus were made into the powder, to observe the effect of hypoglycemic effect on the experimental diabetes model mice.Results found that the pupa infected by recombinant virus can reduce blood sugar more significantly than the normal pupa. These results lay a foundation for how to development of new classes of hypoglycemic drugs and health products.
