家蚕中PGRMC2蛋白的转录、功能分析
Study on the function and transcription of the protein PGRMC2 from silkworm, Bombyx mori
从本实验室构建的家蚕蛹cDNA 文库中,筛选获得一个编码家蚕类孕酮膜受体元件2蛋白的基因序列,基因登陆号为:DQ311390。多序列比对分析该序列编码蛋白发现其可能是细胞内质膜上孕酮受体的一个组分,在调节孕酮参与的细胞内功能方面有重要的作用。为了研究该蛋白在家蚕体内的功能,我们在大肠杆菌Rosetta(DE3)中表 达、纯化了带有His 标签的PGRMC2融合蛋白,并制备了多克隆抗体。通过Western blotting发现PGRMC2蛋白在家蚕五龄幼虫的头、丝腺、生殖腺、脂肪体等组织中均有表达,其中头部、精巢、卵巢中表达量较高,在家蚕发育各时期中,我们发现随着家蚕的发育PGRMC2的表达量也在增加,其中蛾期最高,进一步提取了五龄家蚕各组织及家蚕各发育时期的总RNA,通过荧光定量PCR法,我们发现PGRMC2转录水平在蛾期最高,在卵期很低,并在五龄幼虫中生殖腺最高,中肠最低。表明该蛋白在家蚕的发育和生殖系统中发挥重要作用。
he gene PGRMC2 is a novel gene from silkworm, Bombys mori. Up to now, there is no literature reporting about any characters about this gene in silkworm. We obtained the gene sequence which encoding PGRMC2 protein of Bombyx mori from the silkworm pupa cDNA library constructed by our lab with the GeneBank Accession Number was DQ311390. PGRMC2 contains an open reading predicted molecular weight of 26.05kD. The gene was inserted into a vector pET-28a with both EcoRⅠ and XhoⅠ restriction sites and then expressed in E. coli Rosseta (DE3). The target protein was purified by affinity chromatography and used to immune a male New Zealand rabbit for the production of anti-serum. Total protein of tissues of the fifth instar larva (Bombyx mori) were extracted and analyzed by Western blot. The results showed that PGRMC2 protein widely exist in the tissues of fifth instar larva (Bombyx mori). Meanwhile, the result of RT-PCR shows that there were high transcriptional level in ovary and testis, and higher with the development of silkworm. So we could deduce PGRMC2 played a very important role in the regulation of the growth and development of silkworm. This result laid a good foundation of further studies of PGRMC2.
