目的 研究丙酮酸盐对低血糖诱导的海马神经细胞死亡的影响及可能的保护机制。方法 利用大鼠海马组织切片培养的方法在体外模拟颅内低血糖环境,将实验为空白对照组、低血糖组和丙酮酸盐治疗组,用碘化丙啶染色方法检测海马组织CA1区神经细胞死亡,利用Lowry and Passonneau 免疫荧光法测定海马组织区内ATP水平。结果 对照组比较,低血糖组海马组织CA1区锥体神经细胞死亡显著增多,海马神经细胞内ATP水平也显著降低(P<0.05)。丙酮酸盐治疗组与低血糖组比较,海马组织CA1区锥体神经细胞死亡显著减少,海马神经细胞内ATP水平也显著升高(P<0.05)。结论 丙酮酸盐可明显改善低血糖诱导的海马神经细胞死亡;丙酮酸盐可能通过增加海马组织内ATP的水平来发挥其神经保护作用。

Objective To study the effects of pyruvate on hypoglycemia induced neuronal cell death of hippocampus. Methods The study was divided into control group,hypoglycemia group,and hypoglycemia + pyruvate group. Propidium iodide (PI) method was used to quantify neuronal death of hippocampus. Luciferin-luciferase luminescence assay was used to quantify ATP levels. Results Hypoglycemia group compared with control group showed that the neuronal cell death in CA1 of hippocampus increased significantly and the ATP levels of hippocampus decreased significantly (P<0.05). Pyruvate group compared with hypoglycemia group showed that the neuronal cell death in CA1 of hippocampus decreased significantly and the ATP levels of hippocampus increased significantly (P<0.05). Conclusions Pyruvate could ameliorate hypoglycemia induced the neuronal cell death; pyruvate could preserve the ATP levels to play a neuronal protection.
