卫气月周期性变化与代谢标志物的相关性研究

目的 运用代谢组学方法研究卫气月周期性的物质基础变化,寻找其潜在标志物, 为卫气本质研究开辟一个新途径。方法 以秋分日所在的农历月,确定月初(农历八月初一)、月中(八月十五)及月末(八月三十)。10只大鼠分别在相应日期的正午12:00取血,采用高效液相色谱-质谱联用仪(HPLC-MS)检测血浆代谢物,利用主成分分析(PCA)和偏最小二乘法(PLS)对数据进行统计分析,比较各组之间血浆代谢组学数据,探索各组之间差异性代谢标志物,推测寻找卫气月周期的潜在标志物。结果 与月中比较,月初1-磷酸鞘氨醇、植物鞘氨醇、皮质醇、二十碳五烯酸、鹅去氧胆酸硫酸盐、脯氨酸、富马酸二甲酯等7种差异代谢物为区分两组的可能标志物;月末亚油酰胺、1-磷酸鞘氨醇、磷脂酰甘油、二十碳五烯酸、脯氨酸、皮质醇、富马酸二甲酯、肉毒碱等8种差异代谢物为区分两组的可能标志物。与月末比较,月初胆固醇乙酸酯、脯氨酸、富马酸二甲酯、皮质醇、植物鞘氨醇、肉毒碱、亚油酰胺、二十碳五烯酸、甘氨熊脱氧胆酸等9种差异代谢物为区别两组可能标志物。进而推测二十碳五烯酸、皮质醇、脯氨酸、富马酸二甲酯等4种差异代谢物为月周期变化的潜在差异性标志物。结论 卫气月周期变化可能与脂质代谢、氨基酸代谢密切相关,同时还伴随激素的调节。。

Objective:To use metabonomics method to study the change of the basic material of month rhythm of Wei Qi, and to find the potential markers, as the essence of the Wei Qi theory research opens up a new way. Methods: Chinese lunar calendar in August the first day for early group, the 15th day for medium group, the 30th day for late group. The 10 rats were collected blood at the corresponding date of the 12:00 points.High performance liquid chromatography mass spectrometry instrument(HPLC-MS) was used to detect plasma metabolites, and principal component analysis(PCA)and partial least squares(PLS) was used to make statistical analysis to the data for comparing plasma metabonomics original data of each group,and explore the differences between groups in possible metabolic marker.Meanwhile coniecture potential biomakers of month rhythm of Wei Qi. Results: Compared with medium group, sphingosine-1-phosphate, phytosphingosine, cortisol, eicosapentaenoic acid, chenodeoxycholic acid sulfate, proline, dimethyl fumarate of early group as possible markers are to distinguish between the two groups; Linoleamide,sphingosine-1-phosphate, phosphatidyl glycerol, eicosapentaenoic acid, proline, cortisol, dimethyl fumarate, carnitine of late group as possible markers are to distinguish between the two groups. Compared with late group, cholesteryl acetate,proline, dimethyl fumarate, cortisol, phytosphingosine, carnitine, linoleamide, eicosapentaenoic acid, glycoursodeoxycholic acid of early group as possible markers are to distinguish between the two groups. Speculation eicosapentaenoic acid, cortisol, proline, dimethyl fumaratefor month rhythm potential markers. Conclusion: Month rhythm of Wei Qi may be related to lipid metabolism, amino acid metabolism closely related, and also accompanied by regulation of hormone.
