Ku蛋白主要以Ku70和Ku80 (高等生物为86)二聚体与DNA结合的方式存在于真核生物体内,有修复DNA双链断裂、调节转录以及V(D)J重组等多种生物学功能。Ku与肿瘤的发生、发展关系密切,与肿瘤的放、化疗耐受密切相关。Ku蛋白有可能成为肿瘤早期诊断、评价预后及放化疗敏感性预测的指标,以Ku作为分子靶点可能在逆转肿瘤放化疗耐受的基因治疗中有较好的临床应用前景。

Ku protein is a multifunctional heterodimer of eukaryotes that consists of two subunits: Ku70 and Ku80. The Ku protein plays a key role in multiple nuclear processes, e.g., DNA repair, chromosome maintenance, transcription regulation, and V(D)J recombination. Now the existence of relation between Ku protein functioning and processes that lead to transformation of normal cells into cancerous ones is determined. Ku plays an important role in the sensitivies to radiation and chemotherapy for cancer. Ku might be a new target for the treatment of lung cancer and theoretical foundation on study of proteomics of apoptotic gene in drug-resistance.
