牙周组织健康人群龈沟液内细胞因子表达水平初探
Primary Study of Cytokines Secretion in Gingival Crevicular Fluid
目的：初步探讨牙周组织健康人群龈沟液（Gingival Crevicular Fluid, GCF）内粒细胞集落刺激因子（granulocyte-CSF，G-CSF）、巨噬细胞集落刺激因子（macrophage-CSF，M-CSF）、粒细胞-巨噬细胞集落刺激因子（GM-CSF）、白细胞介素（interleukin，IL）-1β、IL-4、IL-6、IL-10、IL-12(p70)、IL-13、IL-17、干扰素诱导蛋白-10（interferon inducibleprotein-10，IP-10），单核细胞趋化蛋白-1（monocyte chemtactic protein-1，MCP-1），巨噬细胞炎症蛋白-1α（macrophage inflammatory protein-1α，MIP-1α），巨噬细胞炎症蛋白-2（macrophage inflammatory protein-2，MIP-2），干扰素-γ诱导的单核因子（monokine induced by IFN-γ，MIG），调节活化正常T细胞表达与分泌的趋化因子（regulated uponactivation normal T cell expressed and secreted，RANTES）、血管内皮细胞生长因子（vascular endothelial cell growth factor，VEGF）、肿瘤坏死因子（tumor necrosis factor，TNF）-α，干扰素（interferon，IFN）-γ的普遍水平，为后续诊断研究提供分析基础。方法：使用滤纸条法采集牙周组织健康人群每人4-8颗后牙（第二前磨牙及第二磨牙）的龈沟液，通过微量龈沟液炎症标志物组合的Luminex液相芯片多因子检测体系测定细胞因子表达水平。结果：龈沟液内19种细胞因子含量（pg/ml）（?X±Sd）为G-CSF：1181±1329；GM-CSF：107.4±88.08；M-CSF：67.17±99.36；IL-1β：35.15±32.66；IL-4：3.837±3.644；IL-6：149.9±414.4；IL-10：232.2±1149；IL-12(p70)：33.52±25.62；LIF：83.9±106.9；IL-13：1097±1457；IL-17：7.008±6.505；IP-10：435.8±398.8；MCP-1：776.9±1550；MIP-1α：145.6±275.6；MIP-2：311±496.4；MIG：1652±2158；RANTES：61.13±92.58； VEGF：822.4±1797；TNF-α：19.44±42.12；IFN-γ：16.91±32.73。结论：初步检测龈沟液内19中细胞因子的正常表达水平。
Objective: To determine the normal level of granulocyte-CSF(G-CSF), macrophage-CSF(M-CSF), GM-CSF, interleukin(IL)-1β,IL-4,IL-6,IL-10,IL-12(p70),IL-13,IL-17, interferon inducibleprotein-10(IP-10), monocyte chemtactic protein-1(MCP-1), macrophage inflammatory protein-1α(MIP-1α), macrophage inflammatory protein-2(MIP-2), monokine induced by IFN-γ(MIG), regulated uponactivation normal T cell expressed and secreted(RANTES), vascular endothelial cell growth factor(VEGF), tumor necrosis factor(TNF)-α, and interferon(IFN)-γ in gingival crevicular fluid from the health and provide an analysis basis for subsequent diagnosis system. Methods: Gingival crevicular fluid (GCF) was sampled with filter paper strips by intra-pocket method from the health(four to eight premolars or molars each person).The levels of cytokines were determined by Luminex. Results: The content of each cytokine was as follows(pg/ml)(?X±Sd): G-CSF:1181±1329; M-CSF:67.17±99.36;GM-CSF:107.4±88.08;IL-1β:35.15±32.66;IL-4:3.837±3.644;IL-6:149.9±414.4;IL-10:232.2±1149;IL-12(p70):33.52±25.62;LIF:83.9±106.9;IL-13:1097±1457;IL-17:7.008±6.505;IP-10:435.8±398.8;MCP-1:776.9±1550;MIP-1α:145.6±275.6;MIP-2:311±496.4;MIG:1652±2158;RANTES:61.13±92.58;VEGF:822.4±1797; TNF-α:19.44±42.12;IFN-γ:16.91±32.73. Conclusion: The normal levels of cytokines are determined preliminarily.
