拟南芥锌指蛋白GIS亚家族基因ZFP6的克隆和功能分析

锌指蛋白是植物基因组中最大、最复杂的基因家族之一,在植物生长发育过程中起着重要作用。本实验室以前的研究结果表明GLABROUS INFLORESCENCE STEMS (GIS) 基因家族GIS、GIS2、ZINC FINGER PROTEIN 8 (ZFP8) 和ZFP5基因通过整合赤霉素和细胞分裂素信号控制拟南芥表皮细胞形成和发育。本研究克隆和分析了一个新的与GIS基因具有高度同源性的GIS亚家族基因ZINC FINGER PROTEIN 6(ZFP6),利用遗传学和分子生物学技术对它们是否参与调控植物表皮细胞发育进行了初步分析。实验结果表明,过量表达的ZFP6促进茎上各器官表皮毛的形成。 ZFP6基因丧失功能的突变体表现在花的萼片,茎生叶,和侧枝等部位表皮毛数量的显著减少。这些研究结果表明,ZFP6基因是调控拟南芥表皮毛形成的一个重要功能基因。
Zinc finger protein is one of the most complex gene family in the plant genome. They play important roles in the different process of plant growth and development. Our previous studies had showed that the GIS gene family GLABROUS INFLORESCENCE STEMS (GIS), GIS2, ZINC FINGER PROTEIN 8(ZFP8) and ZFP5 control trichome initiation through integration of gibberellin and cytokinin signal in Arabidopsis. By using the genetics and molecular analyses, we characterized and investigate whether the GIS subfamily gene ZFP6 also play a key role in the regulation of plant epidermal cell differentiation in Arabidopsis. Our results showed that overexpression of ZFP6 results in the increase of trichomes on the inflorescence organs. The loss of function ZFP6 mutation exhibited reduced trichome number in sepals, cauline leaves and lateral stems. These findings suggest that ZFP6 is an important transcriptional factor to control trichome initiation in Arabidopsis.
