BMP-2和β-NGF联合应用促进牙周组织再生的实验研究
Effect of human bone morphogenetic protein-2 and beta-nerve growth factor on periodontal regeneration
目的:本研究拟在beagle犬根分叉病变区联合应用两种生长因子BMP-2和β-NGF,评价其在牙周再生过程中诱导产生再生骨的数量和质量,观察两种因子联合应用对骨改建的影响。方法:选用24-36个月龄beagle犬6只,在下颌第2、3、4前磨牙制备感染的III度根分叉病变,然后以可生物降解的凝胶为载体复合BMP-2和β-NGF,根分叉区局部给药。给药八周后,处死动物,以扫描电镜、Ca/P比分析和组织学观察评价再生组织的质量和数量。结果:在所有组别中,BMP-2和β-NGF联合应用组再生的骨组织Ca/P比最高,并且在胶原纤维表面形成了有更多钙化组织的高密度结构。组织形态学分析显示,联合应用组形成的新牙槽骨和牙骨质明显多于空白和阴性对照组。结论:在人工制备的beagle犬III根分叉病变区局部联合应用BMP-2和β-NGF能提高再生骨组织的数量和质量。
his study evaluates the quality and quantity of regenerated bone in periodontal regeneration following topical application of bone morphogenetic protein-2 (BMP-2) and beta-nerve growth factor (beta-NGF) into class III furcation defects. Thirty-six inflamed class III fucation defects were created in six beagle dogs at sites of mandibular premolars 2, 3 and 4, and then biodegradable hydrogel incorporating BMP-2 and beta-NGF was topically applied to the defects. Eight weeks after application, the quality and quantity of regenerated tissue were evaluated by scanning electron microscopy observation, calcium/phosporus tatio analysis, and histologic evaluation. Histomorphometric analysis revealed that 0.4% rhBMP-2 + 2% beta-NGF promoted the highest percentage of periodontal regeneration among all groups. The results of this pilot study suggest that a topical application of BMP-2 and beta-NGF may improve the quality and quantity of regenerated bone in artificially created Class III furcation defects of beagle dogs.
