目的 探讨甘草清热解毒的功效成分,为甘草酸及其复方制剂的质量标准提升、二次开发提供思路和借鉴。方法 基于课题组报道的的天然黄酮的抗菌定量关系,对《中国药典》收载甘草3个品种所含亲脂性黄酮的化学结构信息进行收集、筛选及其抗菌活性计算;同时,对甘草水煎液经乙酸乙酯萃取所得的亲脂性成分进行理化与波谱分析、抗菌活性测定,并结合中医药典籍的佐证和现代文献探讨这些亲脂性黄酮与甘草清热解毒功效的相关性。结果 甘草共计报道约134个亲脂性黄酮,几乎全部为化学结构中含有1~2个异戊烯基单元的异戊烯基黄酮;抗菌活性计算显示这些黄酮具有很强的抗革兰氏阳性菌活性,其中最低抑菌浓度为0.4~40.0μg/mL的化合物占94.0%;同时,甘草水煎液所含的亲脂性成分具有显著抗菌活性,且含有上述可能的异戊烯基黄酮,尤其是异戊烯基二氢黄酮。结论 甘草所含的异戊烯基黄酮是其清热解毒的重要功效成分,并与祛痰止咳的功效关联,可将其纳入甘草质量标志物(Q-markers)的筛选范围,且优选其所含的异戊烯基二氢黄酮;同时,甘草用其清热解毒、祛痰止咳时,以丸入药为佳。

Objective To explore the functional ingredients of Glycyrrhizae Radix et Rhizoma clearing away heat and removing toxic substances, and then to provide ideas and references for the improvement of quality standards, and the secondary development, of this traditional Chinese medicine and its compound preparations. Method: Based on the quantitative antibacterial relationship of flavonoids reported by our group, the chemical structures of lipophilic flavonoids isolated from three species of licorice in Chinese Pharmacopoeia were collected and screened, and then their antibacterial activities were calculated. Simultaneously, the lipophilic ingredients, extracted with ethyl acetate from Glycyrrhizae Radix et Rhizoma decoction, were performed for the physicochemical analysis, spectral analysis and antibacterial test. Finally, the correlation between the function clearing away heat and removing toxic substances and these lipophilic flavonoids, of Glycyrrhizae Radix et Rhizoma, were further explored by the combinational analyses of some evidences of Chinese medicine classics, the research results and the literatures reported. Results: A total of 134 lipophilic flavonoid compounds form Glycyrrhizae Radix et Rhizoma were reported. Almost all of them are isopentenyl flavonoids containing 1 to 2 isopentenyl units. The calculation for the antibacterial activities of these flavonoids showed that they had strong inhibitory activity to Gram-positive bacteria, and those with the minimum inhibitory concentrations from 0.4 to 40.0 μg/mL were accounted for 94.0%. Simultaneously, the lipophilic ingredients in the licorice decoction present antibacterial activity, and contain the above possible isopentenyl flavonoids, especially isopentenyl dihydroflavones. Conclusions: The isopentenyl flavonoids are a class of important ingredients of Glycyrrhizae Radix et Rhizoma for clearing away heat and removing toxic substances, and also related to the efficacy of eliminating phlegm and relieving cough. Thereout, they can be considered as the screening range for the quality markers (Q-markers) of licorice, especially for the isopentenyl dihydroflavones. Furthermore, it is better for Glycyrrhizae Radix et Rhizoma to be ground into powder for preparing pills when it is used to clear away heat and remove toxic substances, and to eliminate phlegm and relieve cough.
