本文合成并表征了三个钌(II)配合物[Ru(bpy)2(biim)]2+ (1)、[Ru(phen)2(biim)]2+ (2) 和[Ru(p-mopip)2(biim)]2+ (3)(bpy是联吡啶,phen是邻菲罗啉,p-mopip是4-甲氧基苯咪唑[4,5-f][1,10]菲罗啉)。采用圆二色谱(CD)、荧光共振能量转移(FRET)和PCR-stop实验方法研究了钌(II)配合物与人端粒DNA序列 5′-G3(T2AG3)3-3′(HTG21)的相互作用。结果表明配合物1、2能诱导人端粒DNA构象发生明显的改变,且三个配合物都能稳定G-四链体结构。

hree ruthenium(II) complexes [Ru(bpy)2(biim)]2+ (1), [Ru(phen)2(biim)]2+ (2) and [Ru(p-mopip)2(biim)]2+ (3) where bpy is 2,2′-bipyridine, phen is 1,10-phenanthroline, biim is 2,2′-biimidazole and p-mopip is 2-(4-methoxyphenyl)-imidazo- [4,5f]phenanthroline), have been synthesized and characterized in this work. The interaction of human telomeric DNA sequence 5′-G3(T2AG3)3-3′(HTG21) with ruthenium(II) complexes was investigated via circular dichroism (CD) measurements, fluorescence resonance energy transfer (FRET) melting assay, polymerase chain reaction (PCR) stop assay. The results indicated that complexes 1, 2 could induce a remarkable conformational change of human telomeric DNA, and three complexes have the ability to stabilize the G-quadruplex.
