左归丸含药血清对大鼠骨髓间充质干细胞骨向分化中Ⅰ型胶原基因表达含量影响的实验研究

Objective：Study on the change of type-I collagen mRNA expression during the differentiation into osteoblasts from rat marrow mesenchymal stem cells induced by zuogui pill medicated serum.Methods Use the method of adherent culture to culture marrow mesenchymal stem cells and then consecutive to purify mesenchymal stem cells. Conventional induction group use β-glycerophosphate, dexamethasone, vitamin C to induce mesenchymal stem cell differentiate into osteoblasts and the experimental group on the basis of conventional induction group with Zuoguiwan medicated serum. Stained using alizarin red staining of calcium nodules and Alkaline phosphatase staining to identify the successful induction. The expression of Type-I collagen mRNA was determined using Real time-PCR.Results Whole bone marrow adherent and differential adhesion method combined with used to get the purified mesenchymal stem cells. The expression of type-I collagen displayed a 16-fold and 3-fold increased at day 21 compared to those in day 7 and day 14 in Zuoguiwan medicated serum group and also were significant higher than those in non- medicated serum group.Conclousin Rat mesenchymal stem cells induced by the drugs could differentiate into osteoblasts,Zuoguiwan induction of serum to promote the expression of type-I collagen mRNA
