三种继代培养基对番茄离体叶片再生能力的影响
Effects of three kinds of subculture medium on the regeneration of tomato leaves in vitro
以番茄S2品种为试验材料,研究了三种添加不同激素配比(0.2mg/L ZT, 2mg/L ZT ,2mg/L ZT +0.1mg/L IAA)的继代培养基番茄离体叶片出愈率和出芽率的影响。结果表明:供试的三种继代培养基对番茄子叶愈伤组织的诱导效果没有差异,诱愈率均可达到100%。其中培养基中含有IAA成分的培养基中叶片出芽率最高,达到32.71%;其次是单加2mg/L ZT的筛选培养基,出芽率为11.54%;单加0.2mg/L ZT的培养基中外植体的出芽率为6.80%。研究结果对离体番茄叶片的再生有一定的参考价值。
In this reseach, the tomato cultivar S2 was used as test materials, and the effection of the leaves in vitro callus rate and the sprouting rate in the subculture medium that included three different hormone combinations (0.2mg/L ZT, 2mg/L ZT, 2mg/L ZT +0.1mg/L IAA) were studied. The results showed that the three kinds of subculture medium had no different effection on the induction of leaf callus induction, and the induction rate reached 100%. Tomato leaves sprouting rate in the culture medium containing IAA component reached 32.71%, which was the highest among these three kinds of culcture medium; followed by screening single plus 2mg/L ZT medium, the budding rate was 11.54%; the bud sprouting rate in the culture medium that plus 0.2mg/L ZT was 6.80%. The research results have a certain reference value for the regeneration of tomato leaves in vitro.
