碘化N-正丁基氟哌啶醇对大鼠缺血再灌注心肌细胞肌浆网钙泵的影响
Effects of N-n-Butyl Haloperidol Iodide on Sarcoplasmic Reticulum Ca2+-ATPase of Rat Myocardium during Ischemia/Reperfusion
目的:研究碘化N-正丁基氟哌啶醇(N-n-butyl haloperidol iodide, F2)对缺血再灌注(ischemia/reperfusion, I/R)大鼠心肌细胞肌浆网钙泵(sarcoplasmic reticulum Ca2+-ATPase, SERCA)活性及蛋白表达的影响。方法:结扎大鼠冠状动脉左前降支制作心脏I/R模型。缺血前舌下静脉给予不同剂量的F2,采用无机磷比色法检测心肌肌浆网SERCA的活性,Western-blot方法检测心肌特异SERCA蛋白的表达。结果:与对照组相比,缺血(30 min~60 min)再灌注(0 min~120 min)不同时程中,心肌SERCA蛋白表达量未发生改变,但活性明显降低,差异具有统计学意义(P<0.05)。缺血前给予不同剂量的F2 (0.25 mg/kg、0.5 mg/kg、1 mg/kg、2 mg/kg 和4 mg/kg)均能抑制I/R所致的SERCA活性下降(P<0.05),且呈剂量依赖性,不同剂量的F2对心肌细胞SERCA蛋白表达量均未影响。结论:F2可剂量依赖性的改善I/R状态下SERCA的活性,这可能是F2具有调节钙稳态进而保护I/R心肌作用的另一重要机制。
Objective: To investigate the effects of N-n-Butyl Haloperidol Iodide (F2) on activity and protein expression of sarcoplasmic reticulum Ca2+-ATPase (SERCA) of rat myocardium during ischemia/reperfusion. Methods: The rat myocardial I/R model was established by occluding left anterior descending coronary artery (LAD). F2 in different dosages was given through the sublingual vein before ischemia. The SERCA activity was tested by inorganic phosphorus colorimetric method. The SERCA protein expression was examined by Western-blot. Results: Compared with the respective Sham Groups, the myocardial SERCA activities of I/R Group (30~60 min ischemia followed by 0~120 min reperfusion) were significantly decreased, but were significantly inhibited in a dose-dependent manner when F2 in different dosages (0.25 mg/kg、0.5 mg/kg、1mg/kg、2mg/kg and 4 mg/kg) was given to rats (P<0.05). The expression levels of SERCA protein in myocardium had no differences among Sham Group, I/R Group and different dosages of F2 Groups. Conclusion: F2 dose-dependently ameliorated the decreased activity of SERCA caused by I/R, which might be another major mechanism for its role in maintaining calcium homeostasis, and thus its cardio protection against IR injury.
