羽状鸡冠花幼苗玻璃花超低温保存研究

超低温保存被认为是目前长期保存种质资源最理想的方法,但以幼苗为保存材料的报道很少。本研究以‘新视野’羽状鸡冠花(Celosia plumosa ‘Fresh Look’)幼苗为材料,研究建立了其玻璃化超低温保存技术程序。具体为:取胚根长至2-3mm的无菌播种幼苗,25℃装载40min,0℃下PVS2脱水60min,更换新鲜的PVS2溶液并迅速投入液氮,最少冻存24h后取出,40℃水浴化冻60s,再用添加400 U?mL-1 CAT的Unloading溶液洗涤两次,每10min换液一次,冻存后幼苗转入恢复培养基暗培养7d后,转入正常光(光照强度约为40 μmol?m-2?s-1)下继续培养,植株再生率可达83.33±1.57%。
ryopreservation is considered to be the best way for long-term germplasm conservation. As few reports about seedling cryopreservation, a procedure has been developed for vitrification of Celosia plumosa ‘Fresh Look’seedlings. The procedure was: seedlings with 2-3 mm radicle length were loaded at 25℃ for 40min, then dehydrated seedlings in vitrification solution (PVS2) at 0℃for 60 min, immersed seedlings into LN2 directly and thawing in a water bath at 40℃ for 60s, following that, washed twice (10min each) with unloading solution added with 400 U?mL-1 CAT. Then recultured vitrified seedlings on MS medium in darkness for 7ds prior to exposure to the light. The regeneration rate reached 83.33±1.57%.
